Lee Donghae merupakan salah satu anggota Super Junior. Dan termasuk salah 1 dari 4 leader dance dari Super Junior. Awalnya Donghae ingin menjadi seorang atlet, namun dibawah pengaruh sang ayah, akhirnya dia memutuskan untuk menjadi seorang penyanyi. Pada tahun 2001, ia berhasil menandatangani kontrak dengan SM Entertainment setelah berhasil memenangkan Best Outward Appearance bersama dengan Sungmin pada SM Youth Best Contest.

Sebelum bergabung dengan Super Junior, awalnya Donghae masuk dalam grup Smile bersama dengan Leeteuk, namun grup ini dibubarkan sebelum melakukan debutnya. Akhirnya pada tahun 2004, Donghae bersama dengan Leeteuk bergabung dengan grup Super Junior. Pada November 2005, beberapa hari sebelum Super Junior melakukan debut resminya, Donghae membuat penampilan singkat dalam acara SBS Shin Dong-yup's There Is There. Pada 6 November 2005, Donghae bersama dengan anggota Super Junior lainnya melakukan debut resmi mereka dalam program musik SBS Popular Song dengan membawakan single pertama mereka "Twins (Knock Out)".

Donghae memulai debut aktingnya dalam film horor dokumenter Super Junior Mystery 6. Pada bulan Agustus 2006, Donghae berperan sebagai manusia robot dalam video musik BoA "Key of Heart" versi Korea. Pada Juli 2007, Donghae bersama dengan anggota Super Junior lainnya, bermain dalam film produksi SM Entertainment, Attack on the Pin-Up Boys. Donghae berperan sebagai teman sekelas sekaligus teman dekat Kim Kibum. Seorang siswa SMA yang menyelidiki serangan misterius pada "pretty boy" di berbagai sekolah tinggi. Pada akhir 2007, Donghae muncul dalam video musik "Kissing You" milik Girls' Generation. Dia juga tampil dalam sebuah iklan "Happy Bubble" bersama dengan Kyuhyun.

Pada April 2008, Donghae bergabung dengan sub grup Super Junior, yang akan melakukan aktivitas di Cina, Super Junior M. Kemudian di tahun berikutnya, Donghae bersama dengan Siwon membintangi sebuah video musik Firefly [萤火虫] milik penyanyi Taiwan, Ariel Lin. Pada bulan Juli, dia menyanyikan lagu ciptaannya sendiri, Beautiful, dalam konser tur mereka, Super Show 2, dan dirilis dalam Super Show 2 Concert Album. Dia juga menciptakan lirik lagu "A Short Journey" bersama dengan Eunhyuk, yang dirilis dalam album repackage Bonamana.

Pada bulan November 2010, Donghae membintangi drama It's Okay, Daddy's Girl sebagai Choi Wook Gi, satu dari tiga bersaudara. Dia juga menyanyikan lagu soundtrack untuk dramanya "Just Like Now" bersama dengan Ryeowook. Pada tahun 2011, Donghae menyanyikan lagu tema untuk 8th Asia Song Festival, "Dreams Come True", dengan Seohyun dari Girls 'Generation. Single ini dirilis pada tanggal 11 Oktober 2011 sebagai single digital. Hasil dari penjualan disumbangkan kepada Unicef ​​untuk membantu anak-anak di negara-negara Afrika.

Tahun 2011, Donghae bersama dengan Siwon membintangi sebuah drama Taiwan Skip Beat!, bersama dengan Ivy Chen. Dia juga menyanyikan lagu soundtrack drama "This is Love" bersama dengan Henry Super Junior M.

Pada tanggal 16 Desember 2011, Donghae & Eunhyuk merilis single digital Oppa, Oppa, di mana mereka pertama kali menyanyikannya di konser Super Junior dari Super Show 4 di Seoul pada 19 November 2011. Single ini juga dirilis di Jepang dengan video musik asli Jepang pada tanggal 4 April 2012.

Bulan Mei 2012, Donghae kembali berakting dalam drama Ms Panda and Mr Hedgehog bersama aktris Yoon Seung-ah. Donghae juga menyanyikan soundtrack drama "Plz Don't", yang dirilis secara online pada tanggal 20 Agustus.
